Because the cell wall of AFB has lipid component, so normal staining does not work. Phenol using for Fluoroscent staining method melts the lipid of cell wall, and get staining by penetrating auramine-rhodamine into cell. Cell wall with Auramine-rhodamine sustains even exposured to decolorizer like acid-alcohol, so it is expressed as AFB.

(+) Test with an AFB QC silde, Bright yellow or green for positive.
(-) Test with an AFB QC silde, Black (background) for negative.
