Because cell wall of AFB contains lipid component, so normal staining method does not work. For phenol using for Ziehl-Neelsen method melts the lipid of cell wall, get staining by penetrating Carbol-fuchsin into cell. Cell wall with Carbol-fuchsin sustains even exposured to decolorizer like acid-alcohol, so it is expressed as AFB.
